摘  要:[目的]以醋酸铅染毒小鼠为模型,研究铅对肝脏超氧化物歧化酶(SOD)活性及CuZn-SODmRNA表达的影响。[方法]分别以浓度为0、0.1%、0.2%、0.4%醋酸铅喂养小鼠。6周后取肝脏,试剂盒测定SOD活性;RT-PCR法测定CuZn-SODmRNA表达水平。[结果]与对照组比较,染毒组SOD活性明显下降(P﹤0.01),CuZn-SODmRNA表达显著下调(P﹤0.01),且两指标均存在剂量-效应关系(r=0.901,P﹤0.01)。[结论]铅可致小鼠肝脏SOD活性下降,CuZn-SODmRNA表达下调,两者存在良好效应关系,呈正相关。[Objective]To study the effect of lead acetate on activity of superoxide dismutase and CuZn-SOD mRNA expression in mice's hepatocytes.[Methods]32 male KM rats were randomly divided into 4 groups.Each group received lead acetate at random in drinking water at a concentration of 0,0.1%,0.2% or 0.4% for 6 weeks,respectively.The activity of SOD in liver was detected,and the expression of CuZn-SOD mRNA was determined by RT-PCR.[Results]Compared with the control group,the activity of SOD and the expression of CuZn-SOD mRNA of experimental groups were both decreased significantly(P﹤0.01),they also presented significant dose-effect relationship(P﹤0.01).[Conclusion]The activity of SOD and the expression of CuZn-SOD mRNA are depressed by lead acetate in rats' liver.
