鞘内注射米诺环素对甲醛诱导的大鼠炎性痛的作用

摘  要:目的观察鞘内注射米诺环素对甲醛致痛大鼠的行为学和脊髓背角c-fos表达的影响。方法 24只雄性SD大鼠随机分为正常对照组、生理盐水组和米诺环素组,每组8只。对照组不做任何干预,生理盐水组和米诺环素组在右足底注射5%甲醛前30 min分别鞘内注射生理盐水20μl、米诺环素50μg。记录足底注射甲醛后1 h内每5 min的舔爪和缩腿时间,然后免疫组化观察脊髓背角c-fos的表达。结果米诺环素鞘内注射明显减少甲醛诱导的缩腿和舔爪时间,显著减少注射侧脊髓背角c-fos的表达(P<0.05或<0.01)。结论米诺环素鞘内注射能减少甲醛诱导的疼痛反应和脊髓背角c-fos的表达。Objective To observe the effect of intrathecal injection of minocycline on formalin-induced nociceptive behaviour and c-fos expression in the spinal dorsal horn in rats.Methods Twenty-four adult male SD rats were randomly divided into three groups with 8 rats for each: the normal control group,saline group and minocycline group.Animalsin the control group did not receive any treatment.Normal saline 20 μl and minocycline 50 μg(20 μl),were given intrathecally 30 min prior to intraplantar injection of formalin(5%,50 μl) to the right hindpaw in saline-and minocycline-treated rats.The total time of licking and flinching in 5 min interval was counted for 1 hour after injection formalin,and then the c-fos expression in the spinal dorsal horn was measured by immunohistochemical method.Results Intrathecal injection of minocycline obviously shortened the time of licking and flinching and significantly decreased the c-fos expression in the ipsilateral spinal dorsal horn induced by intraplantar injection of formalin(P0.05 or 0.01).Conclusion Intrathecal injection of minocycline attenuates formalin-induced pain response and c-fos expression in the spinal dorsal horn.
