胃空肠旁路术对非肥胖型糖尿病小鼠的影响

摘　　要：目的观察Roux-en-Y胃空肠旁路术(GBP)对链脲霉菌素(STZ)所致非肥胖型糖尿病小鼠模型血糖等的影响,血糖与体质量变化的关系及手术的安全性。方法成年雄性C57BL/6小鼠经腹腔注射STZ(200 mg/kg),建立非肥胖型糖尿病模型。成模后小鼠随机分为3组:GBP组(DMOP组)、假手术组(DMFOP组)、未处理组(DM组),另设立正常对照组3组:正常未手术组(N组)、正常假手术组(NFOP)组和正常手术组(NOP)。分别测术前和术后第1、2、3、4、5、6周各组实验动物的随机血糖及体质量变化,观察术后生存情况。结果 (1)与DM、DMFOP组相比,DMOP组的小鼠血糖下降明显,并可维持到术后6周以上(实验观察结束)。(2)GBP后,糖尿病小鼠的体质量不降低。(3)DMFOP组(15/16)与DMOP组(2/16)比较,小鼠围手术期死亡率差异有高度统计学意义(P<0.01)。结论 (1)GBP对非肥胖型糖尿病小鼠具有降血糖作用,且降糖作用与体质量变化无明显相关性。(2)GBP是较为安全的术式。Objective To investigate the effect of Roux-en-Y gastric bypass（GBP） on blood glucose in streptozotocin（STZ）-induced diabetic mice,the relationship between blood glucose and body weight,and the safety of surgery.Methods None-obese diabetic mouse,which were produced with mature C57BL/6 mouse by peritoneal injection of STZ（200 mg/kg）,were allocated into three groups randomly： GBP group（DMOP）,the shame operation group（DMFOP） and the untreated control group（DM）.At the same time,the 3 normal mice control groups were set： GBP group（NOP）,the shame operation group（NFOP） and the untreated group（N）.Blood glucose were measured in all mice before and 1,2,3,4,5,6 weeks after operation and body weight were measured simultaneously.Results（1） Comparing with the DM or DMFOP control group,random blood glucose in mice were reduce significantly post operation.The declination of blood sugar sustained more than 6 weeks after operation（till the end of experimental investigation）.（2） Diabetic mouse body weight didn＇t reduce after GBP surgery.（3） Perioperative mortality was different between DMOP（2/16） and DMFOP（15/16） group.Conclusions（1） GBP can markedly reduce plasma glucose in the none-obese diabetic mice,unrelated to body weight.（2） GBP is a safe surgery for mice.
