摘  要:目的:探讨脑力苏胶囊对小鼠脑组织海马细胞增殖及凋亡的影响。方法:采用双侧颈总动脉结扎反复脑缺血再灌注的方法制备血管性痴呆小鼠模型。模型成功建立后将其随机分为模型组、假手术组、西药喜得镇组、脑力苏高剂量组、脑力苏低剂量组,后3组为治疗组。给药15 d后进行行为学测验,并采用流式细胞术检测各组海马细胞增殖指数及凋亡率的变化。结果:模型组小鼠学习成绩与记忆成绩下降,各治疗组小鼠的学习成绩与记忆成绩均有提高。流式细胞术检测模型组小鼠海马细胞增殖指数及凋亡率明显增高;其中以脑力苏胶囊低剂量组最为明显,各用药组均低于模型组。结论:脑力苏对血管性痴呆小鼠有明显治疗作用,其作用机理可能与抑制海马细胞过度增殖及凋亡有关。Objective:To investigate the effects of Naolisu capsule on proliferation and apoptosis expression in hippocampus cell of mice with vascular dementia. Methods:The bilateral common carotid artery ligation and repeated cerebral ischemia and reperfusion were prepared for mouse model of vascular dementia.Model were successfully established and the mice randomly divided into model group,sham group,Hydergine group,Naolisu capsule high dose group and low dose group;the last three groups were treatment groups.Recorded the behavior of 15 days after the administration of tests,and detected change of apoptosis and proliferation rate in hippocampal cell by flow cytometry. Results:Academic performance and memory were decreased in model group,were improved in treatment groups.Cell proliferation and apoptosis in the hippocampus were significantly higher in model group by flow cytometry;in which treatment groups were obviously lower than the untreated group,especially in low dose group. Conclusion:Naolisu capsule to mice with vascular dementia have significant therapeutic effects,its mechanism may be related to excessive inhibition of hippocampal cell proliferation and apoptosis.
