摘  要:目的在超声条件下提取甘草中有效成分甘草多糖(GPS)并测定其含量,以筛选获得适宜的甘草多糖提取工艺条件。方法采用正交试验设计,选取提取时间、提取温度及料液比作为工艺因素,每个因素采用3个水平,选择L9(34)正交表进行实验,同时用硫酸-苯酚法测定甘草多糖的含量来作为综合评价指标。结果在超声条件下提取甘草多糖的适宜条件为:提取次数为3次,提取时间为20 min,提取温度为45℃,料液比为1∶20(g.mL-1),该提取条件下甘草多糖的含量为2.39%。结论利用超声提取多糖相对其他提取方法有较大优势,可缩短提取时间、提高提取效率,且操作方便、自动化程度高,便于大规模工业生产,为从药材或制剂中提取甘草多糖提供了依据及参考。Objective To extract the effective component,glycyrrhiza polysaccharide(GPS)of glycyrrhiza under ultrasonic condition and determine its content so as to acquire the proper extraction technical conditions of GPS.Methods Orthogonal experimental design was adopted.Extraction time,extraction temperature and solid-liquid ratio were taken as technical factors.3 levels were adopted in each factor.L9(34)orthogonal table was selected in the experiment.Sulfuric acid-phenol method was used to measure the content of GPS which was regarded as the comprehensive assessment index.Results Under ultrasonic condition,the proper extraction technical conditions of GPS:extraction frequency:3 times,extraction time:20min,extraction temperature:45 ℃,solid-liquid ratio:1∶20(g·mL-1).Under these extraction conditions,the content of GPS was 2.39%.Conclusion The ultrasonic extraction of polysaccharide is more advantageous than other method.It may shorten the extraction time and improve the extraction efficiency.It is simple in operation and high in automatic degree.It is suitable for large-scale industrialized production and provides the evidence and reference for GPS extraction from the medicinal material or preparation.
