摘 要:为了研究硫普罗宁(Tiopronin) 对急性肝损害是否有保护作用, 应用D-氨基半乳糖(D-GalN) 造成大鼠急性肝损伤模型。实验分成正常组(N)、模型组(D-GalN)、模型+ 硫普罗宁组(D-GalN+ T)、模型+ 肝得健组(D-GalN+ E)。观察血清中丙氨酸转氨酶(ALT)、肿瘤坏死因子(TNF-α)、肝细胞及血浆中脂质过氧化物(LPO) 和肝组织及红细胞超氧化物歧化酶(SOD) 以及相关的病理变化。结果表明: 模型+ 硫普罗宁组(D-GalN+ T) 及模型+ 肝得健组(D-GalN+ E) 血清中ALT、TNF-α及LPO与模型组相比明显下降(P< 0.01), 而D-GalN+ T和D-GalN+ E两组的SOD活性明显高于模型组(D-GalN) (P< 0.01)。提示: 硫普罗宁能降低血清中ALT、TNF-α的含量及肝组织中氧自由基的产生, 降低血浆和组织中LPO水平, 并能提高细胞SOD活性, 从而减轻肝损伤。其对肝细胞的保护作用与肝得健一致。The protective effect of Tiopronin(T) on D GalN induced acute liver injury was studied. Thirty five rats were divided into normal (N), model group (D GalN), experimental group (D GalN+T), and experimental control group (D GalN+Essential forte). Serum alanine amainotransferase (ALT), TNF α, LPO in hepatic cells, plasma and SOD in erythrocyte and hepatic tissues were determined. The experimental results showed that serum ALT, TNF α, LPO in hepatic cells and plasma concentrations were markedly decreased in D GalN group ( P <0 01). SOD activity in erythrocyte and heptic tissues was obviously increased in the D GalN+T group and D GalN+E group, as compared with D GalN group ( P <0 01). These findings suggest that Tiopronin ould reduce serum ALT, TNF α contents and the production of oxygen free radical, decrease the LPO level in the hepatic cells and plasma and increase SOD activity in cells, so as to alleviate hepatic injury. The protective effect of Tiopronin is similar to that of essential forte.
