摘  要:目的:研究同型半胱氨酸(homocysteine,HCY)对心肌细胞钙摄取的影响及其作用机制。方法:采用同位素技术测定培养大鼠心肌细胞钙的摄取,观察不同HCY浓度对培养心肌细胞钙摄取的影响及蛋白激酶C(proteinkinaseC,PKC)抑制剂或蛋白磷酸酶抑制剂与HCY同时培养对培养心肌细胞钙摄取的影响。结果:HCY呈浓度依赖性促进心肌细胞钙的摄取,HCY促进心肌细胞钙摄取的作用为PKC抑制剂H7和多粘菌素(polymycinB,PB)所抑制,并且受蛋白磷酸酶抑制剂Okadaicacid作用而增强。结论:HCY促进心肌细胞钙摄取,可能与PKC所介导的蛋白磷酸化过程有关。Objective: To study the effect of homocysteine (HCY) on calcium uptake in rat cardiomyocyte and its mechanism. Methods: The calcium uptake by rat cardiomyocyte was assayed with isotopic methods. The in vitro study observed the influences of liner increasing concentrations of HCY, and those of protein kinase C (PKC) inhibitors and selective inhibitor of protein phosphatase type 1 and 2A, Okadaic acid, combined with HCY on the calcium uptake in rat cardiomyocyte. Results: HCY promoted the calcium uptake in a dose dependent way, and this promoting effect was blocked by PKC inhibitors, H7 and polymycin B, but stimulated by selective inhibitor of protein phosphatase type 1 and 2A, Okadaic acid. Conclusion:The calcium uptake in rat cardiomyocytes was induced by HCY possibly through cellular PKC pathway.
