贵州省2个猪种APOA5基因比较分析

摘　　要：根据GenBank中报道的猪载脂蛋白A5(apoA5)基因序列设计3对引物,通过PCR方法分别对可乐猪和贵州白香猪的apoA5基因进行扩增、测序,将2个猪种apoA5基因序列进行比对,并找出基因变异位点。结果表明:可乐猪apoA5基因DNA长为2474bp,白香猪apoA5基因DNA长为2473bp,分别包含3个外显子,2个内含子。2个猪种apoA5基因编码区长皆为1092bp,编码363个氨基酸。白香猪apoA5基因序列与可乐猪相比,存在12个突变位点,其中6个位点为颠换,5个为转换,1个为缺失,两者核苷酸同源性为99.6%。这为进一步研究猪apoA5基因的多态性、生物学活性以及与猪生产性状相关性研究奠定了一定基础。According to pig apolipoprotein A5 （ apoA 5） gene sequence in GenBank, the author designed 3 pairs of primers for amplification of Kele pig and Guizhou White Xiang pig ’ s apoA 5 gene respectively by PCR and sequencing, and compared the two pigs species apoA 5 gene sequences to identify the genetic variation. The results showed： Kele pig apoA 5 gene DNA length was 2474 bp. White Xiang pig gene apoA 5 gene DNA length was 2473 bp, which contained three exons and two introns respectively. Ap oA5 gene coding region in both of the two pig breeds were 1092 bp encoding 363 amino acids. Compared the White Xiang pig apoA 5 sequence with the Kele ’ s, there were 12 mutations, including six transversion, five transition, one deletion mutation, the two pig ’ s nucleotide homology was 99.6%. It provided a certain foundation for the further study of pig apoA 5 gene polymorphism, biological activity and the pig ’ s production traits 。
