摘  要:为了研究CsCCD7蛋白的生物学功能,并为CsCCD7的表达调控提供理论依据,使用一系列生物信息学软件、数据库和在线程序,对黄瓜CsCCD7核酸序列及蛋白序列进行分析。结果表明,CsCCD7开放阅读框长1665bp,编码554AA,与CCD7蛋白同源性高;预测CsCCD7位于黄瓜第6号染色体上、基因全长含7个外显子和6个内含子。CsCCD7分子量是62.7KDa、是不稳定蛋白;其蛋白序列中富含磷酸化位点;此蛋白为非分泌型蛋白、无跨膜结构域;预测其定位于叶绿体;此蛋白二级结构预测表明β折叠和β转角加起来的量多于α螺旋的量、为亲水性蛋白质;其二级结构和三级结构预测显示属于类胡萝卜素双加氧酶家族成员。CsCCD7属于CCDs蛋白家族成员,为黄瓜腋芽生长抑制基因的同源基因,可能在黄瓜腋芽生长和发育阶段对独角金内酯的合成和信号转导途径中起到重要作用,CsCCD7有进一步研究的价值。The aim was to analyze the nucleotide and protein sequences ofCsCCD7,and provide the theoretical bases to understand the biological functions of unknown proteins.Using a series bioinformatics software,database and online programs,the nucleotide and protein sequences of CsCCD7 were analyzed.The results showed that the ORF ofCsCCD 7 consisted of 1665 bp,and encoded a 554 AA protein which was homologous with carotenoid oxygenase 7;CsCCD 7 was located at Chromosome 6 in cucumber,and its full genomic sequence consisted of seven extrons and six introns.CsCCD7 molecular weight was 62.7 KDa and was an unstable protein;there were many phosphorylation sites in CsCCD7.CsCCD7 was non-secretory protein,had no transmembrane domain.CsCCD7 was located in chloroplast,and belonged to CCD7 evolution branch by phylogenetic analysis.CsCCD7 secondary structure contained several kinds of conformations,and the quantity of β-sheet plus β-turn was more than quantity of α-helix,so this protein was hydrophilicity protein.Both secondary and tetary structures’prediction results of CsCCD7 showed it was classified into carotenoid oxygenase family.CsCCD7 was the member of CCDs family,and the homologous gene of the gene of inhibiting the axillary bud growth in cucumber.It may play a role in cucumber branching development,strigolactone’s synthesis and signal transduction pathway.The function of CsCCD7 is deserved to study further.
