金花茶2种体胚发育途径蛋白表达的双向电泳分析

摘  要:本试验在已经建立的金花茶(Camellia nitidissima Chi.)花状多子叶形胚发育途径的体胚循环再生系统基础上,用双向电泳的方法按发育时间顺序分析了金花茶正常体胚和花状多子叶形胚2种体胚发育途径各自不同阶段离体培养材料的蛋白表达差异,以及相同发育阶段2种发育途径的蛋白表达差异。结果表明:随着体胚发育的进程,蛋白种类和总数先增多,在子叶胚阶段蛋白种类和数量最多,之后又逐渐减弱甚至消失,在成苗阶段种类最少;金花茶2种体胚发育途径不同阶段蛋白表达存在明显差异,PD(14.9kD)是金花茶花状多子叶形胚发育途径的特有蛋白,在可分化花状多子叶胚的愈伤组织中,以及从球形胚到成熟胚的各个阶段中都有表达。This experiment based on the established multiple cotyledons and flower-like shape(MCFS) repetitive embryogenesis system inCamellia nitidissima Chi.. The difference in the total proteins expression of the materials during the MCFS pathway and normal developmental pathway was analyzed by 2-DE. The results showed that:along with the growth advancement of somatic embryo,the protein variety and quantity increased first,and reached to the most in the cotyledon embryo stage,afterward gradually weakened,even vanished,and became to least in the seedling stage;the differences were distinct and existed both in every single phase of the same pathway and in the same phases of the two different pathways,PD(14.9 kD) was specific protein in the MCFS pathway,that expressed in the calli which could differentiate MCFS embryo,as well as every stage from the sphere embryo to mature embryo.
