重楼总皂苷对人鼻咽癌细胞CNE-2Z周期及凋亡的影响

摘　　要：目的:探讨重楼总皂苷对CNE-2Z人鼻咽癌细胞周期和细胞凋亡的影响。方法:采用MTT法检测重楼总皂苷对CNE-2Z人鼻咽癌细胞的增殖抑制作用;Hoechst-PI双染、流式细胞仪观察重楼总皂苷对CNE-2Z细胞周期及凋亡的影响。结果:重楼总皂苷能抑制CNE-2Z细胞的增殖,其作用呈明显的时间和剂量依赖性;Hoechst-PI双染显示细胞凋亡特征;重楼总皂苷使CNE-2Z细胞被阻滞在S期;60、90、120μg/mL重楼总皂苷分别作用CNE-2Z细胞24 h后,其凋亡率分别为5.5%±0.22%、7.7%±0.51%、15.5%±0.62%。结论:重楼总皂苷可抑制CNE-2Z细胞的增殖,阻滞细胞于S期,从而诱导细胞凋亡。AIM：To study the effect of total saponins saponins of paris pollyphylla root on the cell cycle and apoptosis of human nasopharyngeal carcinoma CNE-2Z cells.METHODS： The cytotoxic effect of total saponins of paris pollyphylla root on CNE-2Z cells was tested by MTT,and on the cell cycle and apoptosis of CNE-2Z cells was observed by Hoechst-PI double staining and flow cytometric analysis.RESULTS： Total saponins of paris pollyphylla root showed an inhibitory effect on CNE-2Z cells.Hoechst-PI double staining images showed apoptotic morphology change.By FCM methods,CNE-2Z cells treated by saponins were arrested in S phase.And the apoptotic rates of CNE-2Z cells were 5.5% ±0.22%,7.7 ±0.51%,15.5% ±0.62%,treated for 24 h at concentration of 60,90,120 μg/mL,with obvious concentration-dependent relationship.CONCLUSION： Total saponins of paris pollyphylla root could inhibit the growth of CNE-2Z cells,arrested in S phase,then induce the apoptosis of the cells.
