摘 要:目的:研究了培养条件对大叶白麻愈伤组织生长和三萜类化合物积累的影响。方法:以大叶白麻无菌苗下胚轴段为材,在MS+0.2 mg/L 6-BA+0.6 mg/L NAA培养基上诱导愈伤组织并继代,探索愈伤组织继代次数、光周期、培养基碳源和添加物对大叶白麻愈伤组织生长和三萜类化合物积累的影响。结果:(1)愈伤组织继代3次增长倍数最大,三萜类化合物的产量达最大值,分别为7.3倍和0.54%;(2)4种光周期处理中,16 h/d光照下的愈伤组织增长量、三萜类化合物含量最高,分别达到为7.2倍和0.52%;(3)蔗糖比葡萄糖、果糖更有利于愈伤组织的生物量增长和三萜类化合物的积累,以附加5%蔗糖最佳,分别为8.3倍和0.72%;(4)在3种含添加物的培养基中,以添加2 mg/L水杨酸的培养基上愈伤组织增长量最大,三萜类化合物含量也最高,分别为7.2倍和0.68%。结论:通过改变培养条件和培养基成分,可提高大叶白麻愈伤组织中三萜类化合物的产量。AIM:To study the effects of different culture conditions on callus growth and triterpene accumula-tion in callus of Poacynum hendersonii[Hook.f.]Woodson.METHODS:The hypocotyls of aseptic seedlings were cultured on MS medium adding 0.2 mg/L 6-BA and 0.6 mg/L NAA to induce callus growth and the subcul-ture,which were used to detect the proliferation times and triterpenes’content under different subculture times,photoperiod,carbon sources and supplements.RESULTS:(1) The growth rate of callus and triterpenes content reached the maximum values on the third subculture,which were 7.3 times and 0.54 % respectively.(2) The callus cultured in 16 h/d photoperiod was optimal for the callus growth and triterpene accumulation,which reached 7.2 times and 0.52 %;respectively.(3) Sucrose showed the stronger ability to induce callus growth and triterpene accumulation than glucose and fructose,5 % sucrose was optimal for callus growth and triterpene accumulation,which reached 8.3 times and 0.72 %;respectively.(4) Callus cultured on MS media adding with 2 mg/L salicylic acid was the most suitable for the callus growth and triterpene accumulation,which reached 7.2 times and 0.68 % respectively.CONCLUSION: The results suggeste that the modulation of culture conditions and medium composition could improve the triterpene accumulation in callus of Poacynum hendersonii.
