摘 要:目的:比较小柴胡汤超微饮片与传统饮片药效及药理学作用剂量关系,为中药复方超微饮片临床应用提供科学依据。方法:采用小鼠免疫性肝损伤模型观察两种饮片对小鼠肝脏、脾脏、胸腺脏器指数、血清转氨酶活性及肝脏病理改变的影响;采用MTT比色法观察药物对小鼠脾淋巴细胞增殖的影响。结果:超微饮片5.07 g/kg与传统饮片10.14 g/kg剂量均可显著降低模型小鼠血清谷丙转氨酶(ALT)活性(P<0.05);超微饮片5.07 g/kg亦可降低血清谷草转氨酶(AST)活性(P<0.05);超微饮片10.14 g/kg与传统饮片20.28 g/kg剂量均可显著减轻肝损伤程度(P<0.05);小柴胡汤超微饮片5.07 g/kg剂量与传统饮片20.28 g/kg剂量可显著抑制脾淋巴细胞增殖(P<0.05)。结论:小柴胡汤超微饮片临床使用量可减少至传统饮片使用量的1/2~1/4。AIM:To comparatively study pharmacodynamic dose-response relationship between the ultrafine powder(UP) and traditional processing(TP)of Xiaochaihu Decoction for clinical application.METHODS: Up and TP were applied to mouse liver injury model to observe the effect on organ index of liver,spleen,thymus and serum transaminase activity and liver pathological change.The influence of Xiaochaihu Decoction both of UP and TP on proliferation of mouse spleen lymphocytes was analyzed by MTT.RESULTS: UP(dose 5.07 g/kg) and TP(dose 10.14 g/kg)could significantly reduce the activity of serum alanine aminotransferase(P〈0.05);UP(dose 5.07 g/kg) could also significantly reduce the activity of serum aspartate aminotransferase(P〈0.05);UP(dose 10.14 g/kg) and TP(dose 20.28 g/kg) could significantly reduce the degree of liver injury(P〈0.05);UP(dose 5.07 g/kg) and TP(dose 10.14 g/kg) could significantly inhibit the proliferation of spleen lymphocytes(P〈0.05).CONCLUSION: UP could reduce to 1 /2 ~ 1 /4 dosage of TP for clinical application.
