摘  要:目的:研究生脉拆方系列注射液人参皂苷Rb1在Beagle犬体内的药代动力学。方法:采用拉丁方实验设计和交叉试验,应用LC/MS/MS分析方法测定Beagle犬血浆中人参皂苷Rb1浓度,并用DAS ver 2.1软件计算其药代动力学参数。结果:人参皂苷Rb1线性范围为0.002 36~9.45μg/L,方法回收率在73.31%~100.12%,日内、日间RSD值均小于15%。4种生脉拆方注射液(生脉、参麦、参味、红参注射液)中人参皂苷Rb1药代动力学参数t1/2如下:(45.63±11.85)、(40.48±11.76)、(32.31±12.60)和(28.37±8.87)h。结论:该法简便、灵敏、特异,适用于血浆中人参皂苷Rb1浓度测定;4种注射液中人参皂苷Rb1在Beagle犬体内药代动力学行为均符合三房室模型;复方与单方注射液的人参皂苷Rb1的t1/2有显著性差异(P<0.05),表明复方配伍影响其在体内药代动力学行为,显示了药物间相互作用。AIM:To study the pharmacokinetics of ginsenoside Rb1 after iv of Shengmai Injection(Panax gin-seng,Ophiopogon japonicus;Schiasandra chinensis) and its dissected prescription in healthy Beagle Dog.METH-ODS: Latin square experimental design with dose,cross-test.The concentrations of plasma samples collected from the Beagle Dogs were determined by LC-MS/MS.The pharmacokinetic parameters were calculated by DAS ver 2.1 software.RESULTS: Ginsenosides Rb1 linear range fell within 0.002 36-9.45 μg/L,The recovery was 73.31%-100.12%.Days,day RSDs were less than 15%.The pharmacokinetic parameters of ginsenoside Rb1 of 4 kinds of the dissected prescription(Shengmai,Panax ginseng + Ophiopogon japonicus;Panax ginseng + Schiasandra chinensis and Panax ginseng) were as follows:t1/2were(45.63 ± 11.85),(40.48 ± 11.76),(32.31 ± 12.60) and(28.37 ±8.87)h,respectively.CONCLUSION: The method for determining the concentrations of ginsenosides Rb1 is simple,quick,not inferfering,and suitable for the measure of ginsenoside Rb1.Plasm concentration of gin-senoside Rb1 in the Beagle dog pharmacokinetics is fitted with the three-compartment model.Pharmacokinetic pa-rameters t1/2between groups have significant difference(P〈0.05),and show that there are drug interactions.
