摘　　要：目的:应用定量代谢组学研究天芪降糖胶囊(黄芪、天花粉、女贞子、石斛、人参、地骨皮、墨旱莲、五倍子、山茱萸和黄连)对糖耐量受损(IGT)患者嘌呤嘧啶代谢的干预作用。方法:通过高效液相色谱–紫外–串联质谱(HPLC–DAD–MS/MS)测定糖耐量受损患者给药前后血清中9种嘌呤嘧啶类代谢物的含量即尿酸、肌酐、肌苷、肌酸、黄嘌呤、次黄嘌呤、胸苷、尿苷和腺苷,统计分析血糖和几种差异代谢物在对照组、给药组及给药过程中含量的变化,然后应用化学计量学主成分分析法分析给药前后的整体性差异。结果:天芪降糖胶囊给药后,IGT患者餐后2h血糖有不同程度降低,差异代谢物的含量和主成分分析聚类投影图均能明显向正常组转化。结论:天芪降糖胶囊对糖耐量受损患者嘌呤嘧啶代谢具有一定的干预作用。AIM：To observe the effect of Tianqi Capsule（Astragali Radix,Trichosanthis Radix,Ligustri lucidi Fructus,Dendrobii Caulis,Ginseng Radix et Rhizoma,Lycii Cortex,Ecliptae Herba,Galla Chinenesis Corni Fructus and Coptidis Rhizoma） on the cycles of purines and pyrimidines in patients with impaired glucose tolerance（IGT） by quantitative metabonomics.METHODS：Simultaneous determination of nine metabolites including uric acid,creatinine,inosine,creatine,xanthine,hypoxanthine,thymicline,undine and adenosine in serum of patients with IGT by HPLC-DAD-MS/MS before and after medication administration,statistical analysis of the change of several different metabolites in the control group and treatment group and the change in the administration process after combining with clinical information such as blood glucose,and analysis of the overall effect between before and after medication administration by principal component analysis of chemometrics.RESULTS：Two-hour postprandial blood glucose in patients with IGT reduced in varying degrees after administration of Tianqi Capsule,and the level of several different metabolites and the cluster projection of principal component analysis trended to normal.CON-CLUSION：Tianqi Capsule has certain therapeutical effect on purines and pyriwidines metabolism in patients with impaired glucose tolerance.
