HPLC法测定裸花紫珠片中毛蕊花糖苷及连翘酯苷的含量

摘　　要：目的:测定裸花紫珠片中毛蕊花糖苷及连翘酯苷的含量,建立裸花紫珠片具专属性的含量控制方法。方法:采用Kromasil C18(4.6 mm×250 mm,5μm)色谱柱;流动相:甲醇-0.1%醋酸水溶液(40:60);流速:1.0 mL/min;检测波长:334 nm;进样量:10μL。结果:毛蕊花糖苷在0.48～9.60μg(r=0.999 4,n=7)范围内线性关系良好,平均回收率(n=6)为95.94%,RSD为1.54%;连翘酯苷在0.54～10.80μg(r=0.999 7,n=7)范围内线性关系良好,平均回收率(n=6)为98.33%,RSD为1.68%。结论:经过系统的方法学考察,该方法专属性强,重复性好,结果准确,为裸花紫珠片提供了科学、专属的质量控制方法。AIM：To develop a RP-HPLC method for determining verbascoside and forsythiaside in LuohuaZizhu Tablet（Callicarpa nudiflora Hook.et Arn）.METHODS：The contents of verbascoside and forsythiaside were determined by HPLC system with Kromasil C18（250 mm×4.6 mm,5 μm）.The mobile phase was methanol-0.1% acetic acid solution（40：60）.The flow rate was 1.0 mL/min.The detection wavelength was set at 334 nm.The sample volume was 10 μL.RESULTS：The linear range of verbascoside was with in the range of 0.48-9.60μg（r=0.999 4,n =7）,the average recovery（n=6） was 95.94%,RSD was 1.54%.The linear range of for-sythiaside was within the range of 0.54-10.80 μg（r=0.999 7,n=7）,the average recovery（n=6） was98.33%,RSD was 1.68%.CONCLUSION：The results indicate that HPLC method is simple,accurate,and re-producible,thus it could be used as the quality control in the preparation of Luohua Zizhu Tablet.
