摘　　要：目的研究应用PAMAM-D对异种移植用外源基因hDAF转染猪精子细胞的安全性。方法制备PAMAM-D/hDAFcDNA复合物,将PAMAM-D及复合物与处理后猪精子细胞共孵育,经精子质量检测工作站检测孵育后的精子活力和精子畸形率。结果经精子质量检测工作站检测,PAMAM-D和PAMAM-D/DNA复合物对猪精子细胞活力和精子细胞畸形率没有明显影响。结论 PAMAM-D及其复合物对猪精子细胞没有明显毒性,应用PAMAM-D制备异种移植用转基因动物是一种安全的方法。【Objective】 To research on the safety of PAMAM-D（polyamidoamine dendrimers） mediated foreign gene transfection into pig sperm cells for xenotransplantation.【Methods】 PAMAM-D/hDAF cDNA compounds were made.Then PAMAM-D and the compounds were incubated with washed pig sperm cells.Next,the effects on sperm motility and deformity were evaluated through Sperm Quality Inspection Workstation in the different groups.【Results】 PAMAM-D and the compounds had no obvious impact on sperm motility and deformity through evaluation in Sperm Quality Inspection Workstation.【Conclusion】 As a vector,PAMAM-D and PAMAM-D/hDAF cDNA compounds have no cytotoxicity on pig sperm cells.The approach may provide a safe method for the production of transgenic pigs for xenotransplantation.
