摘 要:目的探讨神经肽-1受体(NK-1R)拮抗剂WIN62577对哮喘大鼠气道平滑肌细胞(ASMC)内钙离子浓度的影响及其调节机制,为哮喘气道反应性的治疗寻找新的途径。方法采用OVA吸入法制作哮喘模型。原代培养大鼠ASMC。细胞内钙离子荧光成像系统检测ASMC内钙离子浓度。Real-time PCR检测各组ASMC中SERCA2mRNA(肌浆网钙ATP酶)及IP3RmRNA(三磷酸肌醇受体)表达,组间采用单因素方差分析比较各组间差异,P<0.05为差异有显著性。结果哮喘组ASMC中钙离子浓度较正常组增高;正常组ASMC内钙离子浓度在P物质干预后迅速升高;哮喘组ASMC内钙离子浓度在WIN62577干预后迅速下降。正常组及NK-1R拮抗剂干预哮喘ASMC组中SERCA2mRNA表达与哮喘组比较,差异均有显著性(P<0.05);哮喘组ASMC中IP3RmRNA表达与正常组ASMC比较差异无显著性(P>0.05);NK-1R拮抗剂干预哮喘ASMC组IP3RmRNA表达较哮喘组比较差异有显著性(P<0.05)。结论哮喘大鼠ASMC中SERCA2表达减少,WIN62577通过提高SERCA2及降低IP3R表达从而降低细胞内钙离子浓度改善哮喘气道高反应性。【Objective】 To observe the effect of NK-1R antagonist WIN62577 on calcium concentration in asthmatic rat ASMC and study the mechanism of it,which would benefit to find a new method for asthma treatment.【Methods】 The rats were inhaled OVA to make asthmatic rat model,primary culture rat ASMC,calcium ions fluorescence imaging system was applied to analyze the calcium ion concentration alteration.Real-time PCR detected the expressions of SERCA2mRNA and IP3RmRNA in different groups,ANOVA was applied to compare the difference between the different groups,P 0.05 was considered as the difference significant.【Results】 Calcium concentration in ASMC of asthmatic rat was higher than in normal group.Calcium ion concentration increased after had intervened by substance P in normal ASMC and decreased after intervened by WIN62577 in asthmatic ASMC.The expression of SERCA2mRNA increased in WIN62577 intervened group when compared with asthmatic group,the difference was significant(P 0.05).The expression of IP3RmRNA in normal group had no difference when compared with asthmatic group(P 0.05).In WIN62577 intervened group the expression of IP3RmRNA was decreased when compared with asthmatic group,the difference was significant(P 0.05).【Conclusion】 The expression of SERCA2 decreased in asthmatic rats ASMC,WIN62577 could increase the expression of SERCA2 and decrease the expression of IP3R to decrease the calcium concentration and take part in improving asthma airway hyper-responsiveness.
