摘 要:目的建立反相高效液相色谱法同时测定人血清中茶碱和多索茶碱浓度的分析方法。方法血清样品加入咖啡因为内标,三氯醋酸沉淀蛋白,采用Ultimate XB-C18(250 mm×4.6mm,5μm)色谱柱分离,流动相为甲醇:水=30:70,流速为1.0mL/min,检测波长为273 nm,柱温为30℃。结果茶碱在0.97~48.40mg/L范围内,线性关系良好,r=0.9999;多索茶碱在1.10~54.80mg/L范围内,线性关系良好,r=0.9998。日内精密度和日间精密度良好,茶碱、多索茶碱的低、中、高3种浓度方法回收率分别为6.01%、2.25%、0.86%和5.71%、2.02%、1.07%。结论该法操作简便,结果准确可靠,可满足临床快速监测的需要。【Objective】 To develop a new method for simultaneously determining theophylline and doxophylline in human serum by RP-HPLC.【Methods】 Trichloroacetic acid was used to precipitate the serum protein and the coffine was used as internal standard.The separation was performed on Ultimate XB-C18(4.6 mm×250 mm,5 μm) column with methanol-water(30:70) as mobile phase.The flow rate was 1.0 mL/min.The wavelength for detection was 273 nm.The column temperature was 30℃.【Results】 The calibration curve of theophylline had good linearity between 0.97~48.40 mg/L,r =0.9999,and the calibration curve of doxophylline had good linearity between 1.10~54.80 mg/L,r =0.9998.The intra-day precision and inter-day precision were all good.The average recovery of low,medium,high concentration of theophylline were 6.01%,2.25%,0.86%,and the average recovery of low,medium,high concentration of doxofylline were 5.71%,2.02%,1.07%.【Conclusion】 This method is accuate,rapid,and simple.It can be used as a routine method by TDM.
