摘  要:目的:探讨屋尘螨注射液的体外生物活性及不同批次注射液的相对稳定性。方法:收集60例单纯尘螨过敏的哮喘患者血清,制备屋尘螨血清池,采用放射免疫吸附(RAST)抑制试验方法测定屋尘螨注射液体外生物活性,并比较不同批次间的稳定性;同时采用聚丙烯酰胺凝胶电泳法比较不同批次屋尘螨注射液蛋白组分有无差异;酶联免疫吸附法(ELISA)测定屋尘螨注射液主要致敏蛋白Der p1的含量。结果:体外RAST抑制试验显示屋尘螨注射液能抑制标准屋尘螨抗原与IgE的特异性结合,具有很强的生物活性,3种批次的屋尘螨注射液抑制试验结果相似,稳定性良好;电泳结果提示3种批次的屋尘螨注射液蛋白组分相同,具有良好的一致性;ELISA法检查发现3种批次的屋尘螨注射液主要致敏蛋白Der p1含量相同。结论:屋尘螨注射液具有较强的体外生物活性,并且批次间稳定性好。OBJECTIVE To investigate the bioactivity of dermatophagoides pteronyssinus solution and the relative stability of different batches in vitro. METHODS 60 dust mite allergic asthma patients" sera were collected to prepare an allergic sera pool. Radioallergosorbent test inhibition was performed to test bioactivity in vitro. The relative stabilities of different batches were compared. Polyacrylamide gel electrophoresis (PAGE) was used to compare the protein ingredients of different batches of Dermatophagoides pteronyssinus solutions. Enzyme-linked immunosorbent assay (ELISA) was used to measure the content of major allergenic protein Der p1 of Dermatophagoides pteronyssinus solution. RESULTS RAST results indicate that Dermatophagoides pteronyssinus solution can inhibit specific IgDs binding ability to standard Dermatophagoides pteronyssinus allergen in vitro. The results of three-batches are similar. PAGE and ELISA results shows that the protein ingredients and the content of Der p1 of the three batches of Dermatophagoides pteronyssinus solutions were the same. CONCLUSION Dermatophagoides pteronyssinus solution has a good bioactivity in vitro; the solution is very stable and reliable in clinical use for Dermatophagoides pteronyssinus allergic patients.
