黄芩素与黄芩苷的体外抗骨肉瘤作用的比较

摘　　要：目的探讨黄芩素与黄芩苷对骨肉瘤细胞系U2OS和U2OS/MTX300生长抑制和诱导凋亡的作用,并比较两者间的差别。方法应用MTT比色法,测定黄芩素与黄芩苷对U2OS和U2OS/MTX300细胞株增殖的影响,检测其抑制骨肉瘤细胞的时间、剂量效应;应用形态学观察,测定黄芩素对U2OS和U2OS/MTX300细胞的凋亡作用,并进行细胞周期的分析。结果黄芩素对U2OS和U2OS/MTX300细胞的生长抑制及诱导凋亡的作用明显比黄芩苷强,且呈时间依赖和剂量依赖性。结论黄芩素在体外对骨肉瘤U2OS和U2OS/MTX300的抑制作用明显比黄芩苷强。OBJECTIVE To study the growth inhibitory and apoptosis-inducing effect of baicalein and baicalin on human osteosarcoma cell line U2OS and U2OS/MTX300.METHODS U2OS and U2OS/MTX300 cell line were treated with various concentrations of baicalein and baicalin.Their effect of proliferation was determined by MTT test and morphology observation.Flow cytometry were applied to detect the baicalein-induced and baicalin-inducde apoptosis and analyse the cell cycle.RESULTS Baicalein was more effective on a growth inhibitory and apoptosis-inducing of U2OS and U2OS/MTX300 cell than baicalin.CONCLUSION Baicalein has an strong growth inhibitory and apoptosis-inducing effect on U2OS and U2OS/MTX300 cell than baicalin.
