摘 要:目的采用HPLC法测定人血液和尿液中的倍他米松。方法以乙酸乙酯为萃取剂、泼尼松为内标。用Welchrom C18柱,甲醇-水(60∶40)为流动相,检测波长240 nm。结果倍他米松在血液和尿液中的线性范围为0.1~20μg.mL-1(r2>0.999);检测限均为0.02μg.mL-1;平均方法回收率为91%~104%;日内、日间RSD≤4.6%;萃取回收率≥86.3%。结论所建方法准确、灵敏、简便,适用于血液、尿液中倍他米松的分析。OBJECTIVE To establish an HPLC method for determination of Betamethasone in the blood and urine of human.METHODS Ethyl acetate was uesed as the solvent for liquid-liquid extraction and prednisone was used as the internal standard.Separation was achieved on Welchrom C18(250 mm×4.6 mm,5 μm) column using CH3OH-H2O(60∶40) as mobile phase.The detection wavelength was at 240 nm.RESULTS The linear limits of Betamethasone both ranged from 0.1 to 20 μg·mL-1(r20.999) in the blood and urine.The both limits of detection were 0.02 μg·mL-1.The mean recovery of this method ranged from 91% to 104%.The intra and inter day precision of Betam ethasone were no more than 4.6% and the mean extraction recovery was no less than 86.3%.CONCLUSION The method is simple,accurate,sensitive and suitable for determination of Betamethasone in the blood and urine.
