HPLC法测定黄连解毒汤传统汤剂与配方颗粒中盐酸小檗碱和黄芩苷含量的研究

摘　　要：目的建立HPLC法测定黄连解毒汤传统汤剂与配方颗粒中盐酸小檗碱与黄芩苷含量的方法。方法色谱柱:C18ODS反相色谱柱;流动相:甲醇-乙腈-0.4%磷酸(50:45:5);流速:1.0 ml/min;检测波长:314nm;柱温:25℃。结果盐酸小檗碱、黄芩苷分别在0.05～1.1 mg/ml(r=0.9997),0.06～1.3 mg/ml(r=0.9998)范围内呈良好的线性关系。所测6组黄连解毒汤传统汤剂中盐酸小檗碱、黄芩苷的含量平均值分别为0.789 mg和1.79 mg,6组黄连解毒汤配方颗粒中盐酸小檗碱与黄芩苷的含量平均值分别为1.16 mg和2.10 mg。结论结果表明黄连解毒汤传统汤剂与配方颗粒中盐酸小檗碱和黄芩苷的含量均以后者较高。Objective Established HPLC method for the determination HJD（Huanglianjiedu Decoction）traditional decoction ready for decoction and dispensing granule of berberine and baicalin content of the method.Methods Chromatographic column： C18ODS Reversed-phase chromatography column;Mobile phase： methanol-acetonitrile-0.4%phosphate（50：45：5）;velocity： 1.0 ml/min;detected wavelength： 314nm;column temperature： 25℃.Results Berberine、baicalin show good linear relationship in 0.05～1.1 mg/ml（r = 0.9997）,0.06～1.3 mg/ml（r= 0.9998） respectively.Measured HJD traditional decoction of berberine,baicalin content 0.789 mg and 1.79 mg,HJD dispensing granule of berberine and baicalin content 1.16 mg and 2.10 mg.Conclusion Results show that between HJD traditional decoction and dispensing granule of berberine and baicalin content,the latter are higher levels.
