摘 要:目的:探讨补肾中药天癸方对雄激素致不孕大鼠(ASR)促排卵的作用机制。方法:新生SD雌性大鼠随机分为3组,ASR模型组:9d龄SD雌性大鼠皮下注射丙酸睾丸酮;中药组:ASR模型在81d龄起每日灌服天癸方;对照组:9d龄SD雌性大鼠皮下注射中性茶油。在106d龄左右(动情前期),放免法测定血Δ4-雄烯二酮(Δ4-A)、总睾酮(TT)、游离睾酮(FT)、雌二醇(E2)、胰岛素(Ins)和C-肽(C-P),RT-PCR方法检测胰腺及卵巢雄激素受体(AR)mRNA表达水平,并观察中药治疗前后上述指标的变化。结果:ASR模型血Δ4-A、TT、FT、E2、Ins、C-P均显著高于对照组(P<0.05,P<0.01),胰腺、卵巢的AR mRNA表达水平明显升高(P<0.01);中药天癸方治疗后,排卵率为58%,各项血激素及胰腺AR mRNA表达水平明显下降,与模型组比较有显著性差异(P<0.05,P<0.01),卵巢AR mRNA水平与治疗前相比无明显改变。结论:ASR模型血雄激素水平升高,上调胰腺、卵巢的AR mRNA表达,引起高胰岛素血症和无排卵。天癸方可降低血雄激素和胰岛素,使胰腺AR mRNA表达水平下降,纠正ASR模型的内分泌-代谢失调状态,虽然卵巢的AR mRNA表达水平仍较高,但在正常的激素环境下仍出现了排卵。Objective:To investigate the mechanism of inducing ovulation with kidney tonifying herbs Tiangui Recipe in androgen sterilized rats(ASR).Methods:The experiment new born SD female rats were divided randomly into 3 groups:the ASR model group,the SD rats were injected with testosterone propionate subcutaneous at the age of 9 days;the herb group,ASR were administered with Tian Gui Fang daily at the age of 80 days for two weeks;the control group,SD rats were injected with tea oil subcutenously at the age of 9 days.Around the age of 106 days(proestrous day),all rats were sacrificed.Serum Δ4-andronestedione(Δ4-A),total testosterone(TT),free testosterone(FT),estradial(E2),insulin(Ins) and C-peptide(C-P) were measured by radioimmunoassay(RIA).The amount of AR mRNA in pancreas and ovary was quantitated by RT-PCR.The indexes were measured after Tiangui Recipe treatment.Results:The mean serum concentrations of Δ4-A,TT,FT,E2,Ins and C-P in ASR models were significantly higher than those in control group(P0.05,P0.01).The expression of AR mRNA in pancreas,and ovary elevated significantly(P0.01) in ASR rats as compared with control group.The rate of ovulation in herb group was 58%.After Tiangui Recipe treatment,serum hormones and AR mRNA in pancreas decreased significantly as compared with ASR models(P0.01),but no significant difference was observed of the AR mRNA in ovary.Conclusion:The elevated serum androgen levels in ASR model could enhance the expression of AR mRNA levels in pancreas and ovary,and then induced hyperinsulinemia and anovulation.Serum androgen,insulin and the expression of AR mRNA in pancreas were decreased after Tiangui Recipe treatment.Tiangui Recipe could alter the dysfunction of endocrinology and metabolism in ASR model.Although the AR mRNA levels in ovary kept high,but in the normal hormone environment,the rats still ovulated.
