健脾解毒方诱导裸鼠人胃癌细胞凋亡相关基因表达的研究

摘　　要：目的:研究健脾解毒方对裸鼠人胃癌皮下移植瘤的抑制作用,利用基因芯片筛选其诱导裸鼠人胃癌细胞凋亡相关基因表达,分析其可能的作用靶点。方法:建立人胃癌MKN-45细胞裸鼠皮下移植瘤模型,随机分为4组:生理盐水对照组、替加氟化疗组100mg·kg-1·2d-1、中药健脾解毒方组(40g/kg)、健脾解毒方联合化疗药组(健脾解毒方40g/kg联合替加氟100mg·kg-1·2d-1,每组12只。给药14d后观察各组瘤体质量量和动物生存期,并利用Affymetrix Genechip U133 Plus2.0基因芯片筛选健脾解毒方诱导凋亡相关基因表达。结果:与对照组相比,健脾解毒方组瘤体质量量明显降低(P<0.01),生存期显著延长(P<0.01)。且健脾解毒方联合化疗药组对肿瘤的生长抑制作用明显优于单纯化疗组和单纯健脾解毒方组(P<0.05)。基因芯片分析结果显示,健脾解毒方治疗后表达丰度相差在2倍以上基因的有879个,其中与细胞凋亡相关的基因有358个。结论:健脾解毒方抗裸鼠人胃癌作用,联合化疗药可提高疗效,其抗癌机制与诱导胃癌细胞凋亡有关。Objective：To investigate the effect of Jianpijiedu Decoction on gastric cancer in nude mice and to explore its regulation of genes expression those related to apoptosis.Method：Gastric cancer models in nude mice with human MKN-45 cell were established,and then were divided into 4 groups：NS control group,Jianpijiedu group,Ft-207 group and Combination therapy group randomly,after intragastric administration for 14 days,the tumor growth,the survival time of the mice in each guroup were compared;Induction of expression of genes related to apoptosis was examined with gene chip.Results：Compared to NS control group,tumor weights in Jianpijiedu group were decreased（P0.01）,while the tumors in Combination therapy group were lighter than the other groups（P0.05,P0.01）.The growth of the tumor were inhibited by Jianpijiedu therapeusis treatment with a growth inhibition 57.04%,the inhibitive effect was enhanced when using Jianpijiedu Decoction with Ft-207,the growth inhibition was 83.10%;the survival time of the mice in Jianpijiedu group and Combination therapy group were extended;Compared to NS control group,there were 879 genes were examined to be variance expression,358 of which were related to apoptosis.Conclusions：The inhibition effect of Jianpijiedu therapeusis may be related to the apoptosis induction of cell.
