平肝潜阳方治疗原发性高血压大鼠的实验研究

摘　　要：目的:通过动物实验探讨平肝潜阳方治疗高血压病的中药药理学机制。方法:将40只原发性高血压大鼠(SHR)随机分为5组:模型组,平肝潜阳方高、中、低剂量组及马来酸依那普利对照组,每组8只;观察平肝潜阳方对SHR宏观表征和行为学的影响,测量给药前及给药后1、2、3、4周大鼠血压和心率,于给药4周后,检测大鼠血清一氧化氮(NO)和内皮素(ET)含量。结果:给药4周后,平肝潜阳方和马来酸依那普利对SHR宏观表征和行为学指标的影响表现出不同的特点,平肝潜阳方在改善大鼠急躁易怒、眩晕、头痛等方面具有一定的量效关系;与模型组相比,平肝潜阳方高、中剂量组能显著降低模型动物收缩压(SAP)、舒张压(DAP)和平均动脉压(MAP),减慢心率(P<0.01),2种药物均能显著增加大鼠血清NO含量,降低大鼠血清ET含量(P<0.01),但两者无显著差异。结论:平肝潜阳方不仅能够明显降低血压,而且能够改善大鼠的宏观表征,可能是通过调节血管活性物质而实现的。Objective：To study the traditional Chinese medicine（TCM） pharmacological basis of the compound prescription formula for calming the liver and suppressing yang in treating spontaneous hypertension of animal experiments.Methods：Forty spontaneously hypertensive rats were randomly divided into five groups：model group,the high,middle,low dosage group of the formula for calming the liver and suppressing yang and Enalapril Maleate tablets group,with eight rats in each group.The observation and determination methods of signs and general behavior were established.BP（blood pressure） and heart rate were measured respectivelyby tail-cuff technique before therapy and one week,two weeks,three weeks,four weeks after therapy.The serum content of NO（nitric oxide） and ET（endothelin） will be detected after four weeks therapy.Results：The influence of formula for calming the liver and suppressing yang and Enalapril Maleate tablets on signs and behavioral indexes of SHR is different.The formula for calming the liver and suppressing yang showed hypotensive effect and improvement of the signs and behavioral indexes in irritability,dizzy,headache with the dose-responses relationship.During four weeks＇ treatment,the formula for calming the liver and suppressing yang significantly lowered the SAP,DAP,MAP and heart rate of rats comparing with the model group（P0.01）.Meanwhile,formula for calming the liver and suppressing yang and Enalapril Maleate tablets can significantly increase NO content,decrease markedly the content of ET（P0.01）,but they have no significant different.Conclusion：The formula for calming the liver and suppressing yang can not reduce the blood pressure,but also improve the signs and behavioral indexes of SHR,the effect is probably concerned with regulating the plasma vasoactive substance activity and content.
