摘 要:目的:评价青风藤超临界提取物对青藤碱雷公藤甲素的裸鼠透皮影响。方法:采用超临界CO2提取青风藤挥发油,并用GC-MS进行分析。体外透皮作用实验采用Franze扩散池法;体内采用微透析联用HPLC的方法。结果:青风藤超临界提取物中(z,z)-9,12-十八碳二烯酸甲酯占43.45%、十六碳酸甲酯占13.24%。离体裸鼠皮肤预先用青风藤超临界提取物处理,两种成分的渗透速率(与空白比)和皮肤滞留量均提高;加入到巴布剂基质中的促渗效果不甚明显,但体内实验促渗效果显著。结论:提示5%青风藤超临界提取物可促进青藤碱雷公藤甲素的透皮吸收。Objective:To evaluate the essential oil from Caulis Sinomenii to promote the percutaeous absorption of sinomenine and triptolide in nude mouse skin.Methods:Essential Oils was extrated by Supercritical CO2 and was analysised by GC-MS.In vitro skin permeation experiments were carried out using a Franz-diffusion cell.Microdialysis coupled withhigh-performance liquid chromatography was developed to on-line determine the concentration of sample in vivo.Results:The predominant component from essential oils washeptadecanoic acid(43.45%),followed by methyl ester(13.24%).In vitro nude mouse skin(prtreated with oils)permeation and deposition of sinomenine and triptolide was increased,whereas the enhancement was relatively weak when the oils was added to patch base.In vivo enhancement of essential Oils was significantly.Conclusion:It has suggested that 5% Essential Oils from Caulis Sinomenii could be the penetration enhancer for sinomenine and triptolide.
