摘 要:抗菌肽(antimicrobial peptide,AMPs)是天然免疫系统中的一种小分子多肽,一些具有抗肿瘤活性,毒副反应小,不易产生耐药性。近年来,深入细致的研究已经明确了抗菌肽对肿瘤细胞的杀伤抑制作用及其作用机制。目前,普遍认同的抗肿瘤抗菌肽的作用机制包括以下五方面:(1)在肿瘤细胞膜上形成穿膜孔道,使肿瘤细胞死亡;(2)作用于细胞骨架使其结构紊乱;(3)抑制DNA合成,影响细胞增殖;(4)作用于线粒体引起肿瘤细胞凋亡;(5)影响免疫效应进而杀伤肿瘤细胞。该文就抗菌肽抗肿瘤细胞的作用机制作一综述。Antimicrobial peptides (AMPs) are a class of small peptides widely existing in many organ- isms, some are with broad spectrum of antitumor activities and uneasily to develop drug resistance. In the last decade intensive research has been conducted to determine the role of innate immunity antimicrobial peptides in the killing effect on cancer cells and the antitumor mechanism of antimicrobial peptides. The general antitumor mechanism of antimicrobial peptides is recognized as following: ( 1 ) to kill the tumor cells by forming pore on the membrane and leading depolarization of cell membrane, (2) to disturb the cytoskeletal structure, (3) to inhibit the DNA synthesis, interfere with cell proliferation, (4) to act on mitochondria to induce apoptosis of tumor cells, (5)to kill the tumor cells by affecting the immune response. This paper reviewed the molecular mecha- nism of killing effect of antimicrobial peptides on tumor cells.
