摘 要:目的 比较三种重组合人工骨的成骨能力。方法 在50 只兔的100 个颅骨缺损动物模型中分别植入:①牛脱蛋白骨(b D P B)/牛骨形态发生蛋白(b B M P)/肿瘤坏死因子α( T N Fα);②b D P B/b B M P/碱性成纤维细胞生长因子(b F G F);③b D P B/b B M P/表皮细胞生长因子( E G F);④b D P B/b B M P。植入后1、2、4、6 及8 周行组织学检查及 X 线摄片; 植入后10 及42 天行 35 S和45 Ca 液闪计数及灰重测定。结果 成骨能力:b D P B/b B M P/ T N Fα> b D P B/b B M P/b F G F( P< 0.01) > b D P B/b B M P/ E G F( P< 0.01);b D P B/b B M P/ E G F 与 b D P B/b B M P 无显著性差异( P> 0.05)。结论 T N Fα与 B M P及载体复合后,在体内对骨修复产生明显的促进作用,在载体/骨诱导因子/生长因子的重组合人工骨模式中, D P B/b B M P/ T N Fα是一种有价值的骨移植材料。Objective To compare the osteogenesis of recombination artificial bones, which are bovine deproteined bone (bDPB) and bovine bone morphogenetic protein (bBMP), combined with tumor necrosis factor α(TNFα),basic fibroblast growth factor (bFGF), epidermal growth factor (EGF) respectively.Methods One hundred trephined skull bone defects in fifty rabbits were divided into four groups, which implanted with bDPB/bBMP/TNFα,bDPB/bBMP/bFGF, bDPB/bBMP/EGF, and bDPB/bBMP respectively.X ray and histological changes were observed in the 1st, 2nd, 4th, 6th, 8th weeks after implantation.The content of 35 S and 45 Ca and ash weight were measured at 10 and 42 days after operation.Results The osteogenesis of bDPB/bBMP/TNFα group was stronger than that of bDPB/bBMP/bFGF group( P<0.01) , while bDPB/bBMP/bFGF group was stronger than that of bDPB/bBMP/EGF( P<0.01) .No significant statistical difference were found between bDPB/bBMP/EGF and bDPB/bBMP( P>0.05) .Conclusion TNFα combined with bBMP and carrier can stimulate bone formation and increase the volume of new bone in vivo .It suggests that bDPB/bBMP/TNFα is a valuable biomaterial of bone graft.
