摘 要:目的:探讨栀子豉汤改善胰岛素抵抗的作用及机制。方法:采用小剂量链脲佐菌素加高脂饲料方法建立Wistar大鼠2型糖尿病模型,观察栀子豉汤对胰岛素抵抗大鼠空腹血糖(FPG)、糖化血红蛋白(HbA1c)、血清胰岛素浓度(FINS)水平的影响,逆转录聚合酶链式反应(RT-PCR)检测肝脏组织中胰岛素受体(InsR)mRNA的表达水平,放射免疫法检测肿瘤坏死因子α(TNF-α)水平。结果:栀子豉汤可不同程度降低空腹血糖、血清胰岛素、糖化血红蛋白,显著上调肝脏InsR mRNA表达,降低肿瘤坏死因子α水平,增加胰岛素敏感指数。结论:栀子豉汤可改善胰岛素抵抗,作用机制可能与其上调InsR mRNA表达水平、降低肿瘤坏死因子α水平有关。Objectives : To investigate the effect and mechanism of Gardeniae and Sojae Praeparati decoction on improving insulin resistance.Methods :The male healthy Wistar rats were given a low dose streptozotocin by peritoneal injection and fed with high calorie laboratory caw to establish the murine model of type 2 diabetes.Observe the effects of FPG,HbA1c,FINS in IR rats.InsR mRNA expression was evaluated by RT-PCR,and TNF-α were measured by radio-immunity.Results :The level of FPG,HbA1c,FINS in Gardeniae and Sojae Praeparati decoction group obviously reduced(P0.01 or P0.05),the expression of liver InsR mRNA significantly increased(P0.01 or P0.05),the level of TNF-α obviously reduced(P0.01).Conclusions :Gardeniae and Sojae Praeparati decoction could improve insulin resisance,the mechanism might be closely correlated with its up-regulating the liver InsR mRNA expression and reducing the level of TNF-α.
