摘 要:目的:探讨多种病理染色条件下,柴胡总皂苷醇洗脱精制品对大鼠肝毒性肝脏损伤病理组织学的影响,为柴胡肝毒性病理学定性研究提供组织学依据。方法:分别采用常规HE染色、脂肪特殊染色和马松染色的方法,通过光镜下阅片观察和病理图像系统分析,观察柴胡总皂苷醇洗脱精制品对大鼠肝毒性肝脏损伤病理组织学改变的影响。结果:柴胡总皂苷醇洗脱精制品不同剂量组对大鼠肝毒性常规HE染色肝组织病理改变可见肝细胞嗜酸性变、肝细胞玻璃样变、炎细胞浸润、肝索结构变化、肝细胞坏死;脂肪特殊染色肝组织病理改变可见呈片状或群集分布的脂肪变性;马松染色肝组织病理改变可见胶原纤维延伸明显,并互相连接包绕整个肝小叶,导致正常肝小叶结构破坏,假小叶形成。药物高、中、低剂量之间的肝毒性损伤程度有一定的剂量依赖关系。结论:连续15 d灌服一定剂量的柴胡总皂苷醇洗脱精制品可导致大鼠明显的肝脏病理组织改变,柴胡总皂苷是其肝毒性主要物质基础。Objective: To investigate the influence on rats' hepatical injury caused by refined products of Bupleurum saikosaponin by alcohol elution in histopathologics based on a variety of pathological conditions,in orde to provide the pathological accordance for the hepatoxicity of Bupleurum.Methods: To observe the influence of refined products of Bupleurum saikosaponin by alcohol elutionon histopathologics of rats' hepatical injury through the light microscope and analyze pathological image analysis system using conventional HE staining,special staining method of fat cell and Mass staining respectively.Results: The pathological changes of conventional HE staining hepatic tissue caused by different dose refined products of Bupleurum saikosaponin by alcohol elution were seen that :acidophilia degeneration of hepatic cells,hyaline degeneration of hepatic cells,infiltration of inflammatory cells,structural changes of hepatic cord and hepatic cells necrosis;the pathological changes of hepatic tissue by special staining method of fat cell were seen that the flaky or clustered distribution of fatty degeneration;the pathological changes of hepatic tissue by masson staining method were seen that the significant extention of hepatic collagen fibers and surrounding the normal hepatic lobular by connecting with each other,which lead to the destruction of normal hepatic lobular structure and the formation of false hepatic lobular.The hepatoxical damage degrees had some dose dependence among the high,medium and low dose,and it is the obvious discrepancy compared with distilled water control group.Conclusion: The refined products of Bupleurum saikosaponin by alcohol elution(81.9%) administrated continuously to rats for 15 days under the certain dose can induce obvious hepatic histopathological changes.The total Bupleurum saikosaponin was the mainly material basis of hepatoxicity.
