摘  要:目的:观察TNBS法UC模型3、7、14天大鼠结肠粘膜ITF、MUC2、TGF-α的变化及中药复方溃结灵对其的影响,并探讨其作用机制。方法:采用TNBS法UC大鼠模型,RT-PCR检测大鼠结肠粘膜ITFmRNA的表达,免疫组化方法观测MUC2、TGF-α的变化。结果:模型大鼠结肠粘膜3、7天ITFmRNA相对表达量明显低于正常组,14天时则与正常组无明显差异;模型大鼠结肠粘膜MUC2、TGF-α表达3天有所降低,7天逐渐升高,14天明显高于正常组。与模型组相比,溃结灵组和SASP组3、7、14天ITFmRNA相对表达量、MUC2蛋白表达均明显增高,而TGF-α表达呈现逐步增高过程,仅在14天时显著高于模型组。结论:溃结灵有促进TNBS法UC大鼠模型结肠粘膜ITF基因、MUC2蛋白及TGF-α表达上调的作用,这可能是其抗UC作用的机理之一。Objective: To observe dynamic changes of ITF,MUC2 and TGF-α in colonic mucosa of ulcerative colitis rat and the effect of Kuijieling Decoction.Methods: Ulcerative colitis model of rat was induced by TNBS.The expression of ITF mRNA was detected with RT-PCR.Immunohistochemistry was performed for MUC2 and TGF-αin colonic mucosa of rats.Results: ITFmRNA expression of model group in the third day and the 7th day decreased obviously compared to normal group,but no sinificant difference in the 14th day.the expression of MUC2 and TGF-αreduced in the third day,and then incresed in the 7th and 14th day.Compared to the model group,ITF and MUC2 expression of the treatment group(KuiJieling decoction and SASP)heightened obviously in the third day,the 7th day and the 14th day,but TGF-α expression increased only in the 14th day.Conclusion: The expressions of ITFmRNA,MUC2 and TGF-α in colonic mucosa of UC rats induced by TNBS increased significantly after treated by Kuijieling Decoction,which could be its partial mechanism of treatment.
