摘 要:目的:观察新型壬苯醇醚(NP-9)膜体内、外杀精作用和家兔抗生育效果。方法:①体内、外杀精试验:进行体外和家兔阴道内不同浓度杀精剂的杀精试验,观察其的最低有效浓度。②家兔抗生育实验:按照在动情期投入不同的药物,将家兔随机分成3组,空白组不给药物与雄兔自然交配,阳性对照组在给予市售NP-9膜10 min后与雄兔交配,实验组在给予新型NP-9膜后立即与雄兔交配,计算妊娠率和半数有效量(ED50)。结果:①新型NP-9膜组溶液中NP-9体外最低有效杀精浓度为0.35 mg/ml,NP-9膜组溶液中NP-9和NP-9原料组体外最低有效杀精浓度均为0.25 mg/ml NP-9,新型NP-9膜体内杀精最低有效浓度为1 mg/kg NP-9。②家兔妊娠率空白对照组为100%,10 mg/kg NP-9阳性对照组为0%,1 mg/kg NP-9实验组为0%。实验组ED50为1.1771 mg/kg,低于阳性对照组(3.7931 mg/kg),P<0.01。结论:新型NP-9膜体外最低有效杀精浓度比壬苯醇醚膜组高,但体内杀精效果高于NP-9膜;而ED50低于NP-9膜,显示新型NP-9膜有良好的应用前景。Objective:To observe the spermicidal action in vitro and in vivo and the antifertility effect of the new type NP-9 membranes in rabbits.Methods:1) The spermicidal test in vivo and in vitro was adopted to examine the lowest effective concentration of the spermatocide.2) The antifertility effect of rabbits was observed using the stochastic method,rabbits were divided into the blank control,the positive control(nonoxinol pellicles,NP-9) and experimental groups(new type NP-9 membranes) according to put different drugs into the vagina inthe estrum.The animals of the blank control and experimental group naturally mated with male rabbits in theestrum without/with the use of new type NP-9 membranes.The animals of the positive control mated with malerabbits at the 10 min after the use of nonoxinol pellicles in the market.According to the number of pregnancy tocount the pregnancy rates and median effective dose(ED50).Results:1) The lowest effective spermicidalconcentration of NP-9 in the new type NP-9 membranes in vitro was 0.35 mg/kg,while that in nonoxinol pelliclesand NP-9 raw material in vitro was 0.25 mg/kg.The lowest effective spermicidal concentration of NP-9 in the new type NP-9 membranes in vivo was 1 mg/kg.2) The pregnancy rate was 100% in blank control,0% in the 10 mg/kg NP-9 positive control group,and 0% in the 1 mg/kg NP-9 experimental group.The ED50 of experimental group was1.177 1 mg/kg lower than that of positive control group(3.793 1 mg/kg)(P0.01).Conclusion:The lowest effectivespermicidal concentration in the new type NP-9 membranes in vitro is higher than that in the nonoxinol pellicles and thekilling sperm effect in vivo is higher.The results show good application perspectives of the new type NP-9 membranes.
