香椿叶提取物的体外抗肿瘤活性

摘　　要：目的研究香椿叶不同提取部位的体外抗肿瘤作用。方法用乙醇对香椿叶进行提取,再用石油醚、氯仿、乙酸乙酯和正丁醇依次对乙醇提取部位进行分段萃取,设鬼针草提取物各萃取部位的不同浓度组、抗癌药顺铂为阳性组和空白对照组,采用四甲基偶氮唑盐(MTT法)比色法,观察香椿叶提取物的各萃取部位对人胃腺癌SGC-7901细胞和白血病细胞K562增殖的抑制作用。结果香椿叶提取物的各萃取部位在体外对SGC-7901和K562有明显的抑制作用,乙酸乙酯部位的半数抑制浓度分别为168.47μg/mL和102.53μg/mL。结论香椿叶提取物的各萃取部位能抑制人胃腺癌细胞SGC-7901和白血病细胞K562的增殖,有较强的体外抗肿瘤活性。Objective To investigate the anti-tumor effect of different extracts of Toona sinensis in vitro.Methods The sample was extracted by ethanol,then the ethanol extract was extracted with petroleum ether,chloroform,ethyl acetate and n-butanol successively.SGC-7901 and K562 cells were treated with different extracts with different concentrations served as the experimental group,treated with C-DDP served as the positive group,and given no treatment served as the blank control group.The inhibitory effects of different extracts of Toona sinensis on proliferation of SGC-7901 and K562 cells were detected by MTT assay.Results The extracts of Toona sinensis induced significantly growth arrested in SGC-7901 and K562 cells which cultured in vitro,and IC50 of these cells which received ethyl acetate extracts for 48 hours were 168.47 μg/mL,102.53 μg/mL respectively.Conclusion The extracts of Toona sinensis can inhibit the proliferation of SGC-7901 and K562 cells,and has obvious anti-tumor activity in vitro.
