摘  要:目的探讨7肽诱导小鼠树突状细胞免疫应答机制。方法采用"Bulk培养法"制备树突状细胞,经7肽免疫10d的小鼠,取脾细胞,分选CD4+T细胞与经过7肽负载并成熟树突状细胞共孵育24h,收集上清,应用流式细胞技术检测上清细胞因子。结果细胞表面CD11c的表达率超过80%,具有典型的树突状细胞形态。实验组CD4+T上清中TNF-α降低,IL-5、IL-2及INF-r均升高(P<0.05)。结论利用此法能在体外培养出大量的未成熟树突状细胞,7肽能诱导以Th1细胞为主的细胞免疫。Objective To study the polypeptide 7-induced immune response of dendritic cells(DC) in mice and its mechanism.Methods DC were cultured with the "bulk" culture method.Splenocytes were collected from the mice 10 days after subcutaneous immunization with polypeptide 7.CD4+ T cells were isolated from splenocytes by cell sorting and incubated for 24 hours.Cytokines in supernatants were detected by flow cytometry.Results The expression rate of CD11c on surface of DC was over 80%.The CD11c showed a typical morphology of DC.The level of TNF-α was lower while the levels of IL-5,IL-2 and IFN-r were higher in supernatant of CD4+ T cells from experiment group than from control group(P0.05).Conclusion A large number of immature DC can be cultured in vitro with the "bulk" culture method.Peptide 7 can induce immune response of Th1 cells.
