摘 要:改进微生物学实验教材上介绍的Schaeffer-Fulton氏芽胞染色法。将涂片的初染用水浴蒸汽加热和烘箱加热,通过显微镜观察不同温度下不同染色时间的染色效果。以5%孔雀绿为染色剂,100℃蒸汽加热4m in或80℃蒸汽加热6 m in能达到好的染色效果。改进方法操作简单易行,加热过程不需添加染料,更符合环保要求;将改进的染色方法运用于微生物学实验教学,收到了良好的教学效果。chaeffer-Fulton's spore staining method introduced in microbiological experiment teaching material was improved after heating the firstly stained smear by water bath steam and oven,the staining effects at different staining temperature and time were investigated through microscopic observation.The results showed that the best staining results could be achieved with 5% malachite green dye,heating by water steam at 100 ℃ for 4 minutes or at 80 ℃ for 6 minutes.This improved method was easy to operate and met the environmental protection requirement since no extra dye needed during the heating process.This method was put into microbiological experimental teaching,and achieved a better teaching results.
