女贞子超临界CO_2萃取物急性毒性及30d饲养试验的研究

摘　　要：将女贞子超临界CO2萃取物给40只,18～22g昆明小鼠以11、13、15g·kg-1剂量一次灌胃,连续观察7d,未发现中毒症状和死亡。按急性毒性分级标准判定,该受试物属无毒级。女贞子超临界CO2萃取物以0.75、2.25、3.75g·kg-1剂量给Wistar大鼠连续饲喂30d。结果表明,各组动物在给药期均存活,并且采食量、体重、外观特征、行为活动、粪便性状均正常;血液学检查,血液生化指标检测,主要脏器重量和脏器系数均在正常范围内;病理学检查观察到高剂量组和对照组无明显差异。结果显示,女贞子超临界CO2萃取物属于无毒级药物,可以做为饲料添加剂使用。In the acute peroral toxicity test,the supercritical CO2 extract of Ligustrum lucidum was investigated in 40 mice with a body weight ranging from 18 to 22 g by pour stomach administration at doses of 11,13,15 g·kg-1.After 7 d,the treatment was tolerated and no mortality occurred.It was considered to be the no-observed-adverse-effect level following the acute toxicity grading criteria.30 d feeding test was investigated in Wistar rats by feeding for 4 weeds.The results showed that no animal died,and feed intake,body weight,physical characteristics,behavior activity,fecal characteristics were all normal;Hematological examination,blood biochemical test,the main organ weight and organ coefficient were within the normal range;the pathological examination observed that high-dose group and the control group were not significantly different.Therefore,the result of our studies indicated safety of supercritical CO2 extract of Ligustrum lucidum and supported the use of supercritical CO2 extract of Ligustrum lucidum as a feed additive.
