摘 要:目的了解酶联免疫吸附试验(ELISA)在鼠疫监测中的优势及特性,对其在鼠疫监测现场应用做出评价。方法对2006年鼠疫监测收集的鼠脏器及血清(鼠、犬、人)分别检测鼠疫菌F1抗原及F1抗体。结果分离培养、RIHA、胶体金、ELISA、PCR法检测鼠脏器中F1抗原的检出率均为阴性;血清利用IHA、胶体金、酶标法检测鼠疫F1抗体的阳性率分别为1.35%(9/667)、0.76%(2/262)、5.04%(33/655),显示酶标法阳性率高于IHA和胶体金法。结论酶标法与IHA比较其Ka值为0.42,呈中度一致;胶体金与酶标、胶体金与IHA比较均呈轻度一致,其K a值分别为0.11、0.19。ELISA具有敏感性高、特异性强、操作简便、结果易于判断等优点,在鼠疫监测中应用前景广阔。Objective To characterize enzyme-linked immunosorbent assay(ELISA) and assess its potential application in plague surveillance in Guangxi.Methods ELISA was performed to detect Yersinia pestis F1 antigen and anti-F1 antibody in rat organs and serum specimens collected under the surveillance system of plague in 2006.Results F1 antigen in rat organs was not detected by the following assays including microbiologic culturing,reverse indirect hemagglutination assay(RIHA),immunochromatographic assay(ICA),ELISA and PCR.The prevalences of Anti-F1 antibody in serum detected by IHA,ICA.and ELISA were 1.35%(9/667),0.76%(2/262) and 5.04%(33/655).respectively,suggesting that the positive rate obtained by ELISA was higher when comparing with IHA or ICA.Conclusion Medium consistence was observed with a Ka value of 0.42 when comparing ELISA with IHA,a conventional assay.Slight consistence was found when comparing ICA with ELISA or ICA with IHA,with the Ka values of 0.11 and 0.19,respectively.ELISA is highly specific and sensitive,and easy to perform and read.Thus,it is a useful tool for plague surveillance.
