摘 要:目的:筛选川芎、前胡中具有视神经保护作用的有效成分并探讨其视神经保护作用的机理。方法:采用胰酶消化法将出生23 d的SD乳鼠视网膜制成细胞悬液,接种于经多聚鸟氨酸和层粘连蛋白包被的96孔板中,培养2 d后分别加入不同浓度的阿魏酸、川芎嗪、欧前胡素、异欧前胡素、白花前胡甲素、紫花前胡苷溶液继续培养3 d,采用MTT比色法测量各组的吸收值;同法制备细胞悬液,接种于经多聚鸟氨酸和层粘连蛋白包被的24孔板中,培养3 d后分别加入经筛选的川芎嗪、白花前胡甲素、欧前胡素、异欧前胡素溶液,除空白对照组外,其余各组均在80 mmHg体外高压下继续培养2 d,采用Caspase-3,Fas蛋白免疫组化染色检测以及原位末端脱氧核苷酸转移酶标记法检测其Caspase-3、Fas蛋白表达变化以及细胞凋亡情况。结果:体外常压下与空白对照组比较,阿魏酸、川芎嗪、白花前胡甲素、欧前胡素以及异欧前胡素给药组的吸收值明显增加(P〈0.05,P〈0.01);体外高压下与模型对照组比较,川芎嗪、白花前胡甲素、欧前胡素给药组的Caspase-3阳性表达指数、Fas阳性表达指数以及细胞凋亡指数以及细胞凋亡指数明显降低(P〈0.05,P〈0.01)。结论:川芎嗪、白花前胡甲素、欧前胡素以及异欧前胡素均能促进体外常压培养的视网膜神经细胞存活,前三者还能抑制体外高压诱导的视网膜神经细胞节的凋亡。Objective:To study the neuroprotectiveeffecteand mechanismsof rhizoma chuanxiong and radix peucedani.Methods:Retinacell suspensionwas made fromSD suckling mice after born 2 or 3 daysby pancreatic enzyme digestion,and was inoculated into the 96 cell covered by ornithin and laminin.After 2 days,different concentrations offerulic acid,chuanxiongzine,imperatiorin,isoimperatorin,praeruptorin B and nodakeninwere added and cultivated 3 days.The absorption value was measured byMTT.Prepare the cell suspension with the same method and inoculate it into the 24 cellcovered by ornithin and laminin,different concentrations offerulic acid,chuanxiongzine,imperatiorin,isoimperatorin,praeruptorin B and nodakeninwere added and cultivated 3 days.Except the blank group,the rests were cultivated cosntinually for 2 days out-of-boy under the 80 mmHg pressure.in vitro TheCaspase-3、Fas protein expression and the apoptosisindex weremeasured by immunohistochemistry and TUNEL,respectively.Results:Under in ritro normal pressure,comparedwith blank group,the absorption value of ferulic acid,chuanxiongzine,praeruptorin B,imperatiorinandisoimperatorin group wereincreased obviously(P〈0.05,P〈0.01).Under the out-of-body high pressure,comparedwith the model group,all of the expression of Caspase-3,Fasproteinand the apoptotic index of the chuanxiongzine,praeruptorinB,imperatiorin groupwere decreased significantly(P〈0.05,P〈0.01).Conclusion:Chuanxiongzine,praeruptorinB,imperatiorin and isoimperatorincan all promote the survival of retinal nerve cells in normal pressure,and the first three medicines can inhibit the apoptosis ofretinal ganglion cells induced by the high pressure.
