摘  要:目的:研究抗肿瘤抗生素C1027 对人结肠癌HCT8 细胞凋亡相关基因表达的影响。方法:利用cDNA 排列(cDNAarrays)、Northern 杂交、RTPCR 检测技术。结果:用10 nmol·L- 1 C1027 处理HCT8 细胞8 h 后,利用cDNA 排列检测,发现C1027 可以促进TRIP,TRAF3 ,DR4 ,MCH6 ,MCH4,Apo3 ,DR5,ABLL,STAT1 等基因的表达,抑制rhoC的表达。用Northern 杂交及RTPCR 也得到了同样结果。结论:C1027 可能通过调节TNF 受体家族有关的凋亡信号传导通路,诱导细胞凋亡。AIM: To study the effcet of C1027 on the expression of genes related to apoptosis in HCT 8 human colon cancer cells. METHODS: HCT 8 human colon cancer cells were treated with C1027(10 nmol·L -1 ) for 8 hours. The effect of C1027 on the expression of genes in HCT 8 cells were detected by cDNA arrays, Northern blot and RT PCR. RESULTS: Hybridization of the entire cDNA populations to Atlas Arrays showed that C1027 up regulated the expression levels of TRAF3, DR4, DR5, MCH4, MCH6, TRIP, Apo 3, ABLL and STAT1 and down regulated the expression levels of rhoC. These changes were confirmed by Northern blot and RT PCR. CONCLUSION: Most of the genes are involved in TNF receptor family related signal transduction. The results indicate that C1027 may induce apoptosis by regulating TNF receptor family related signal transduction.
