摘  要:【目的】获得一个快速、简洁的提取杏花粉RNA方法。【方法】以新疆杏品种托乎提的花粉为试材,通过3种提取方法和2种材料处理,提取花粉总RNA,并利用电泳和核酸蛋白仪检测其纯度和浓度。【结果】改良的方法M提取花粉得到的RNA产率高,电泳条带整齐、清晰,测得A260/A280在1.8~2.0,反转录后的cDNA可扩增出清晰的特异条带。【结论】该方法既快速简洁,又可获得高质量的RNA,同时也满足花粉中特异表达基因的克隆和鉴定的需要。【Objective】To obtain a fast and simple method for extracting RNA from Prunus armeniaca pollen.【Method】The pollen of "Tuohuti" cultivars of Xinjiang are used as test materials.Three extracting methods and two treatments are established to isolate total RNA of pollen,and detect the purity and concentration by utilizing agarose gel electrophoresis and nucleic acid and protein apparatus.【Result】The bands of the isolated RNA are regular and clear by improved method M.The A260/A280 value of total RNA are between 1.8-2.0, the cDNA after reverse transcription can get amplification of the specific bands of SFB.【Conclusion】This method is not only fast and simple,but also can obtain high grade RNA,simultaneously it satisfies the requirement for clone and identification for the specific gene in pollen.
