摘 要:【目的】镰孢菌是土壤真菌中一类能引起植物和昆虫病害的病原,同时又是可利用的资源。掌握库尔勒地区土壤镰孢菌种类情况,对进一步了解和利用镰孢菌具有深远意义。【方法】运用稀释平板法从土壤中分离到3株镰孢菌,采用形态学及其培养特征进行观察描述,同时利用ITS rDNA序列对其系统发育学进行分析,确定分类地位,并分析比较了基于形态学和ITS rDNA序列分析的两种鉴定手段。【结果】3株菌分别鉴定为芳香镰孢菌(Fusarium redolens)、增殖镰孢菌(Fusarium proliferatum)和木贼镰孢菌(Fusarium equisetic)。【结论】与传统方法相比,ITS rDNA方法有着较高的灵敏度。在对镰孢菌的鉴定中,只有将形态学与分子生物学鉴定相结合,才能使镰孢菌的鉴定分类更加完善。【Objective】The Fusarium is a kind of pathogenic fungi in the soil,which can cause plants and insects disease,and it is also a kind of utilizable resources.It is very important to understand the situation of the Fusariurn species in Kuerle region for further utilization.【Method】The three Fusarium spp.in the soil in Kuerle region was isolated by dilution plate technique and the three fungi strains were identified by the internal transcribed region rDNA(ITS rDNA)sequence analysis and the traditional taxonomy methods.Meanwhile,the two methodologies were compared.【Result】The results showed that three strains were Fusarium redolens,Fusarium proliferatum and Fusarium equisetic.【Conclusion】Compared to taxonomy methods,ITS rDNA identification has higher sensitivity.It suggested that combination of traditional taxonomy methods and ITS rDNA identification could make the Fusarium spp.identification more perfect.
