摘 要:根据NCBI中已注册的近缘树种的SOC1同源基因序列设计引物,采用RT-PCR技术在4年生白桦茎尖中分离得到1条SOC1-like cDNA序列,命名为BpSOC1。该序列编码区长660 bp,编码220个氨基酸,蛋白质相对分子质量为25 367.77 D,理论等电点为9.32。蛋白结构预测分析表明,BpSOC1具有典型的MADS-box结构域和K-box结构域,同时具有多处DNA结合位点、糖基化位点和磷酸化作用位点,属于MADS家族转录因子。系统发育分析表明,BpSOC1属于MADS-box家族的SOC1/TM3亚家族。采用实时定量PCR技术研究不同树龄白桦BpSOC1从5月初至9月在茎尖的时序表达规律,结果显示:BpSOC1无论在2年生苗期的营养生长阶段还是进入3、4年生的生殖生长阶段均有表达,表达高峰均在7月初和9月初。但2年生苗期的BpSOC1表达量最高的7月初和9月初间无显著差异,而进入生殖生长期后9月初的BpSOC1表达量显著高于7月初的表达量,表明Bp-SOC1既参与白桦的营养生长,又参与生殖生长。A SOC1-like cDNA sequence named BpSOC1 was isolated from shoot tips of 4-year-old Betula platyphylla in July by RT-PCR technology,with the primers designed on homologous genes from other closely related species.The gene is 660 bp in length,encoding a protein of 220 amino acid residues with the molecular mass of 25 367.77 D and theoretical pI of 9.32.Protein structure prediction indicates that BpSOC1 is a transcription factor of MADS-box family,which has typical MADS-box domain,K-box domain,multiple DNA binding sites,glycosylation sites and phosphorylation sites.Phylogenetic analysis shows that BpSOC1 belongs to SOC1/TM3 subfamily of MADS-box superfamily.The expression pattern of BpSOC1 in the shoot tips of B.platyphylla of different ages was further investigated using real-time quantitative PCR.Results showed that the expression of BpSOC1 was both in the vegetative growth stage of 2 years old and the reproductive growth stage of 3 or 4 years old,and exhibited two peaks in early July and early September.The highest expression of BpSOC1 from 2-year-old B.platyphylla had no significant difference between early July and early September.When the reproductive growth began,the expression of BpSOC1 in early September was significantly higher than that in early July,indicating that the BpSOC1 involves not only in vegetative growth but also in reproductive growth of B.platyphylla.
