麦门冬汤合苇茎汤抑制小鼠Lewis肺癌生长的体内实验研究

摘　　要：目的研究麦门冬汤合苇茎汤(简称金方/JIN)对Lewis肺癌小鼠移植瘤的抑制作用并初步探讨药物作用机制。方法皮下注射LLC(Lewis lung carcinoma,Lewis肺癌细胞)于C57/BL6小鼠右腋下,观察小鼠生存期、抑瘤率、胸腺指数和肿瘤组织EGFR/Raf/MEK的表达。结果金方可以显著延长小鼠的生存时间,与对照组相比有显著性差异(P<0.01),金方、DDP组对荷瘤小鼠的抑瘤率分别为35%、89%,金方组胸腺指数明显升高,DDP组胸腺指数明显下降,金方、DDP组的EGFR表达均下降(P>0.05,P<0.01),而各组Raf和MEK表达无显著性差异。结论金方可以显著延长荷瘤小鼠的生存期,有一定的抑制肿瘤增长和增强免疫能力的作用,其机制可能与其下调肿瘤组织中EGFR的表达进而影响其下游信号通路的传导相关。OBJECTIVE To investigate the effect of JIN Fang（Maimendong Decoction and Weijing Decoction,are ancient formulae from Golden Chamber,and abbreviated as JIN Fang） on inhibiting Lewis xenografted tumor and the mechanism.METHODS Lewis lung carcinoma cells were inoculated subcutaneously into right axillary of C57 BL /6 mice,and then the following data were observed：the mice survival time,tumor inhibiting rate,thymus index,and the expression of EGFR/Raf/MEK in the tumor tissue.RESULTS JIN could significantly prolong the survival time,（compared with the control group,P0.01）,and the tumor inhibiting rate of JIN and DDP were 35%,89% respectively.JIN significantly increased the thymus index,while DDP group significantly decreased the thymus index.The expression of EGFR in the tissue of mice tumor was reduced in JIN and DDP groups（P0.05,P0.01）;the expression Raf and MEK in different groups had no notable difference.CONCLUSION JIN can not only significantly prolong the survival time of the mice,but also has certain effect on inhibiting tumor growth and enhancing immunity.This may be due to down-regulating the expression of EGFR in tumor tissue and its downstream signal transduction pathway.
