摘 要:目的观察丰白散对慢性阻塞性肺疾病(COPD)模型大鼠肺组织中基质金属蛋白酶-9(MMP-9)及基质金属蛋白酶组织抑制剂-1(TIMP-1)基因表达的影响,探讨其治疗COPD的作用机制。方法取10只大鼠为正常组,不予任何处理,另取大鼠36只采用烟熏加气管内滴加脂多糖(LPS)方法复制COPD模型,取造模成功的30只大鼠随机分为模型组、沐舒坦组和丰白散组各10只,予相应干预28天后,用苏木素—伊红(HE)染色评估各组大鼠肺组织病理改变,逆转录聚合酶链式反应(RT-PCR)检测MMP-9、TIMP-1 mRNA表达水平。结果正常组肺泡结构完整,肺泡间隔无缺损;模型组肺泡结构破坏、融合、弹力纤维网破坏;沐舒坦组、丰白散组病理改变程度明显减轻。模型组大鼠肺组织MMP-9、TIMP-1 mRNA表达水平明显高于正常组(P<0.05);沐舒坦组、丰白散组大鼠肺组织MMP-9、TIMP-1 mRNA表达水平较模型组下降(P<0.05),2组间差异无统计学意义(P>0.05)。结论 MMP-9、TIMP-1过度表达,MMP-9/TIMP-1失衡在COPD气道重构中发挥重要作用;丰白散能下调MMP-9、TIMP-1 mRNA表达,调节MMP-9/TIMP-1失衡,干预气道重构。Objective To observe the effect of the traditional Chinese medicine Fengbaisan on the expression of matrix metalloproteinase-9(MMP-9)and tissue inhibitor of metalloproteinase-1(TIMP-1 ) in rats with chronic obstructive pulmonary disease(COPD),and to investigate the potential mechanisms of the decoction.Methods There were 10 rats enrolled as control group,with another 36 rats induced with COPD model,which was established by modified method of combining fumigation and lipopolysaccharide(LPS) intra tracheal dripping.The 30 successful COPD rats were divided into model group, ambroxol treatment group and Fengbaisan treatment group.After 28 days interventional therapy,the lung tissue sections stained by HE were observed to study the morphological changes,the reverse transcription polymerase chain reaction(RTPCR) was used to evaluate the mRNA levels of MMP-9 and TIMP-1.Results In control group,the alveolus structure was integrities,and no defect in alveolus septum.The typical pathological changes of emphysema were emerged in three groups of COPD model of rats,included the alveolus structure was ruptured and fusion,and elastic fibers was damaged.The pathological changes were released in ambroxol and Fengbaisan treatment groups.Compared with the normal control group,the expression of MMP-9 mRNA and TIMP-1 mRNA in COPD model group were increased significantly(P〈0.05).The expression of MMP-9 mRNA and TIMP-1 mRNA in the Fenbaisan and ambroxol treatment group were decreased markedly as compared to the COPD model group(P〈0.05),while no statistical difference was identified inter-groups(P〉0.05).Conclusion The traditional Chinese medicine Fengbaisan decoction could reduce the mRNA levels of MMP-9 and TIMP-1,and correct the MMP-9/TIMP-1 imbalance,which may be one of the mechanisms of the decoction to treat COPD.
