摘 要:在碱性介质中,高锰酸钾能将糖皮质类激素(醋酸泼尼松和地塞米松)氧化,而本身被还原成绿色的锰酸钾,基于这一反应,本文提出了同时测定醋酸泼尼松和地塞米松的化学计量学-动力学分光光度法。研究发现反应体系在610nm处有一个吸收峰,实验以该波长为检测波长,优化实验条件。在该条件下,检测醋酸泼尼松和地塞米松的线性范围分别为0.8~8.0mg/L和0.02~0.20mg/L,检出限分别为0.4371mg/L和0.0107mg/L。将该方法应用于药物和尿样中醋酸泼尼松和地塞米松的测定,结果与高效液相色谱(HPLC)法无显著性差异。A highly sensitive,selective and simple kinetic method has been developed for the determination of two kinds of glucocorticoids residues,such as prednisone and dexamethasone.The method was based on the reduction of violet potassium permanganate by glucocorticoids in the alkaline medium at 35℃ to form green potassium manganate peaked at 610 nm.Furthermore,linear calibration graphs for each glucocorticoid were obtained in the concentration ranges of 0.8~8.0 mg/L and 0.02~0.20 mg/L for prednisone and dexamethasone,respectively.The detection limits of the proposed method were 0.4370 mg/L and 0.0107 mg/L.The high sensitivity and selectivity of the proposed method allowed its successful application to real samples,and the results were well comparable with those of HPLC method.
