摘 要:目的探讨盐酸氨溴索(Ambroxol Hydrochloride,Amb)联合左氧氟沙星(Levofloxacin,LFX)对粘液型铜绿假单胞菌生物被膜(biofilm,BF)的影响。方法以铜绿假单胞菌粘液型菌株PDO300为实验菌株,构建体外BF模型。分为生理盐水对照组、Amb组、LFX组、Amb与LFX联合作用组,二倍稀释法测定LFX最低杀菌浓度(minimum bactericidal concentration,MBC),结晶紫染色法定量测定BF形成量。结果 PDO300培养7d后可形成稳定的BF。与单独应用LFX相对比,Amb与LFX联合作用后测定载体表面BF值显著降低(P<0.01),且载体表面BF随着LFX浓度增高而减少。结论 Amb可显著增强LFX对BF的破坏作用。Objective To investigate the effect of ambroxol hydrochloride(Amb) combined with levofloxacin(LFX) on biofilm of mucoid Pseudomonas aeruginosa in vitro.Methods Mucoid strains of pseudomonas aeruginosa PDO300 was for biofilm formation in vitro,Minimum bactericidal concentration(MBC) of levofloxacin(LFX) was measured by double dilution.After treated by Amb alone and together with LFX,The biofilms of mucoid Pseudomonas aeruginosa was measured quantitatively with crystal violet staining method with optical density(OD) as index.Results Thick and Stable biolfilms formed after PDO300 was incubated for 7 days.When treated with LFX combined with Amb,The OD value of biofilms of pseudomonas reduced obviously compared with LFX or Amb affected alone(P0.01).Conclusion Amb can significantly enhance the destruct effect of LFX on biofilm of mucoid Pseudomonas aeruginosa in vitro.
