摘 要:利用峰漂移法微乳液电动色谱,评价了相对淌度在药物脂水分配系数(lg Po/w)测定中的应用.使用5种不同疏水性化合物验证了此法的可行性,与采用淌度的测量结果进行了比较;并应用提出的方法测定了3种药物的lg Po/w值,实验值与文献值的平均误差为0.18,准确性好.结果表明,使用相对淌度描述被分析物的迁移行为,有效避免了实验过程中溶液粘度变化、电压、电流以及温度等实验条件的波动引入的误差,提高了lg Po/w测定准确性,为药物疏水性的评价提供一种新的选择.This work evaluates the use of relative mobility to estimate octanol-water partition coefficients (lg Po/w) of drugs using peak-shift microemulsion electrokinetic chromatography (MEEKC). The lg Po/w of 5 compounds with different hydrophobicity were determined by the use of relative mobility and compared with the results obtained by the use of absolute mobility. Furthermore, the developed method was applied to determine the lg Po/w values of 3 drugs, and the average error between the literature value and value determined by relative mobility was 0.18. The result shows that the errors caused by the change of the viscosity and the fluctuation of applied voltage, current and temperature in the experiment could be avoided by the use of relative mobility to describe the analyte migration in MEEKC. The proposed method was an alternative approach to estimate the hydrophobicity of drugs.
