摘 要:目的 探讨姜黄素对支气管哮喘(简称哮喘)大鼠血清、支气管肺泡灌洗液(BALF)中IL-5 及肺组织中 Eotaxin、NF-κB表达的影响.方法 将36只大鼠随机分为正常对照组、哮喘组、姜黄素组,用卵白蛋白(OVA)制备哮喘大鼠模型,正常对照组用0.9%氯化钠溶液代替OVA进行致敏和激发.以ELISA法检测各组大鼠血清及BALF中IL-5的浓度以及计数BALF中嗜酸细胞(EOS)的绝对值及百分比,同时以免疫组化方法检测Eotaxin、NF-κB等在肺组织中的表达.结果 血清及BALF中IL-5浓度的改变与NF-κBp65、Eotaxin蛋白表达的改变:哮喘组及姜黄素组均显著高于正常对照组(P<0.01),姜黄素组显著低于哮喘组(P<0.01).肺组织细胞NF-κB p65的表达与EOS绝对计数、IL-5的表达及Eotaxin的表达均呈显著正相关(r=0.928、0.905、0.850,均P<0.01).结论 姜黄素能抑制NF-κB的活性,通过抑制NF-κB向核内转移与DNA的结合而减少IL-5及Eotaxin的合成,从而发挥抗哮喘作用.Objective To investigate the effects of curcumin on IL-5 levels in serum and bronchoalveolar lavage fluid (BALF), and expression of NF- K B and eotaxin in lung tissue of asthmatic rats Methods Thirty six rats were randomly divided into three groups: normal control group (group A), asthma group (group B), curcumin treatment group (group C). The asthma model was induced by ovalbumin (OVA) challenge. The concentrations of IL-5 in serum and BALF were measured by sandwich ELISA, EOS cells in BALF were counted,the expressions of NF- κB p65 and eotaxin were determined by immunehistochemistry techniques. Results The concentrations of IL-5 in serum and BALF of group B was significantly higher than those of group A and group C (P〈0.01). Immunohistochemistry showed that the expression of NF-κB p65 of was in cytoplasm and nuclei of epithelial cells and inflammatory cells. The expression of NF-κB P65 in group B was significantly higher than those of group A and group C (P〈 0.01). Immunohistochemistry showed that eotaxin was mainly in cytoplasm of the bronchus epithelial cells. The expression of Eotaxin in group B was significantly higher than those of group A and group C (P〈 0.01). The expression of NF- κB p65 in lung tissue was positively correlated with EOC count, IL-5 level and eotaxin expression (r=0.928, 0.905, 0.850,all P〈 0.01 ). Conclusion Curcumin can significantly down-regulate NF- κB and in turns inhibit IL-5 and eotaxin expression in asthmatic rats.
