摘 要:目的:观察豫医无毛小鼠(YYHL)胸腺组织学及淋巴细胞亚群的变化。方法:采用病理组织学和流式细胞术观察2个月龄和6个月龄YYHL和昆明小鼠(n=10)胸腺结构及淋巴细胞亚群改变。结果:6个月龄YYHL胸腺湿质量、胸腺指数、皮质比例、CD4+CD8-、CD8+CD4-/CD4+CD8-分别为(33.80±1.64)g、(0.74±0.05)%、(38.12±1.85)%、(8.85±0.83)%和(0.49±0.02)%,低于6个月龄昆明小鼠的(72.50±4.63)g(t=23.572,P<0.001)、(1.75±0.09)%(t=18.018,P<0.001)、(63.10±3.82)%(t=30.217,P<0.001)、(13.83±1.63)%(t=8.802,P<0.001)和(0.61±0.02)%(t=2.999,P=0.017)。病理组织学观察,6个月龄YYHL胸腺明显萎缩,皮质变薄,淋巴细胞稀疏,髓质区扩大。2个月龄YYHL和2、6个月龄昆明小鼠胸腺组织无显著病理学改变。结论:无毛基因突变诱发了小鼠胸腺萎缩,这可能是造成YYHL寿命短的重要原因。Aim:To explore the morphologic and subsets changes of the thymus in YYHL mice.Methods:The structure and lymphocytes subpopulations of thymus in YYHL mice and Kunming mice(n=10) were observed by histopathology and flow cytometry.Results:The weight,growth index,the percentage of cortical area,CD4+CD8-,and the ratio between CD8+CD4-and CD4+CD8-cells were(33.80±1.64) g,(0.74±0.05)%,(38.12±1.85)%,(8.85±0.83)%,(0.49±0.02)% in 6 month YYHL mice,respectively,which were lower than(72.50±4.63) g(t=23.572,P0.001),(1.75±0.09)%(t=18.018,P0.001),(63.10±3.82)%(t=30.217,P0.001),(13.83±1.63)%(t=8.802,P0.001)and(0.61±0.02)%(t=2.999,P=0.017) in 6 month Kunming mice.Histological examination showed cortical atrophy,which had a disappearance of lymphocytes from the thymic cortex in the thymus of 6-month-old YYHL mice when compared with Kunming mice of the same age.Conclusion:The results suggest degeneration and accelerated age-dependent regression of the thymus in mutant mice,which could explain at least in part the immune defects and shorten lifespan in YYHL mice.
